Title: Manufacturing Three-Dimensional Mini/Microfluidic Platform.
Abstract
Our objective in this study was the development of a novel fabrication process for the creation of 3D mini/microfluidic devices as well as to identify factors that influence the quality of the resulting devices. In this study, a 3D printer was used to create mold inserts of acrylonitrile butadiene styrene (ABS) and the mold inserts were used for casting transparent PDMS microfluidic devices with embedded microchannels. The ABS within the mold inserts was then dissolved using acetone solution. The major challenge of this fabrication process was ensuring the complete removal of the ABS from the microchannels. In the end, we determined that this problem could be largely overcome by sonication-assisted solvent dissolution process. Microfluidic devices, including a tree-like microfluidic device to evaluate the fabrication process and a spiral 3D micromixer, were created for experiments. Experiment results revealed the following: (1) The proposed fabrication process can be used to create 3D microfluidic devices of high complexity, while retaining a high degree of transparency for optically quantification purpose; (2) Sonication is an efficient method by which to facilitate the dissolution of ABS within embedded microchannels; (3) The proposed solvent vapor polishing greatly reduces surface roughness and improves the transparency of the resulting PDMS microfluidic device. With this fabrication technique, the complicated and fully-transparent artificial organ can be achieved in the future for experiments with quantitative results.
