Cancer Biology in Microfluidics
Abstract
In the talk, two major works in our laboratory will be shared. The first topic is “Impedimetric monitoring of cellular response in three-dimensional (3D) environment”. Cell biologists suggested in vitro 3D cell culture provides a more biologically relevant microenvironment, yielding to more physiologically relevant cellular responses. Especially, cancer cells suspended in 3D hydrogel are cultured to proliferate to cancer cell colonies (or tumor spheroids). Conventionally, quantification of the colonies under the tested conditions is achieved by manual counting under microscope. In contrast, we utilized impedance measurement technique to quantify the colonies suspended in the hydrogel. Real-time and automatic quantification of the chemo-sensitive responses of the colonies was objectively obtained for the study of the efficacy of anti-cancer drug. The second topic is “Integration of cell culture and immunoassay on a paper substrate”. In cancer research, investigations of cell signaling and cellular crosstalk are important for understanding the communications that govern cellular activities. However, conventional operations (e.g., cell culture, cell lysis, protein extraction, and immunoassay) for these studies are tedious and time-consuming. We have realized the lab-on-paper concept to integrate cell culture and immunoassay on a paper substrate. Cells were cultured in the paper and cytoplasmic molecules could be directly detected without transferring from one vessel to another one. Studies of cell signaling cascades and cellular crosstalk were demonstrated. This novel analytical tool provides a simple approach for the study of protein expressions under tested conditions.
